I'm pondering Ruby's line, the "it always had to be you". "Always" implies predestiny, as in, before the Special Children contest.

So this is my theory. I think that the Winchesters, as a UNIT, had this destiny - to bring about Lucifer's freedom AND his destruction. They have both been essential to the entire thing - first, Dean made sure Sam won the Special Children Olympics by bringing him back from the dead, but this same action condemned him to hell. What did he do in hell? He broke the first seal and thus started us on the road to armageddon. Then Sam obviously had the role of killing Lilith and releasing Lucifer, and it seems Dean's (and most likely, Sam's with him) destiny is now to defeat Lucifer. Basically, they are inextricably intertwined and both have been essential for the whole shebang. That's why, say, Jake by himself wouldn't have worked.

In short, the Winchesters' love will both doom and save the world. THIS IS SO AWESOME.
